What is ANA Pattern?

The ANA pattern is the microscope staining appearance seen when antinuclear antibodies are detected, and it offers an extra clue about which specific autoimmune condition may be worth exploring further.

When an antinuclear antibody (ANA) screening test comes back positive, laboratories often report the fluorescent staining pattern seen on the cell nucleus, such as homogeneous, speckled, nucleolar, centromere, or peripheral. Each pattern is loosely associated with different underlying antibodies and, in turn, different connective tissue conditions such as lupus, Sjögren's syndrome, systemic sclerosis, or mixed connective tissue disease. The pattern alone does not identify a specific diagnosis; it helps guide clinicians toward which follow-up antibody tests, such as anti-dsDNA, anti-Ro, or anti-centromere, may be most useful. Because ANA staining patterns can also appear in healthy individuals, especially at low titers, the pattern is always considered alongside titer, specific antibodies, and clinical symptoms rather than on its own.
